Personalized tumor models of dynamic colloidal hydrogels

Personalized tumor model study of dynamic colloidal hydrogel is a kind of method using proteins extracted from surgically removed tumors as antigen, combined with dynamic crosslinked hydrogels as carrier, to build an in vitro model that can simulate tumor microenvironment and immune response to evaluate personalized tumor treatment strategies.

A personalized tumor vaccine (DCHVax) based on dynamic covalent hydrogel for postoperative tumor management. Protein extracted from the excised tumor was used as antigen, CpG as adjuvant, and multiarm pegylated (8-arm PEG) / oxidized dextran (ODEX) dynamic cross-linked hydrogel as matrix. Subcutaneous injection of DCHVax can recruit DCs to sites and induce a strong tumor-specific immune response. Thus, it effectively inhibited the growth of postoperative residual tumors in several mouse tumor models.

The application of hydrogels in tissue engineering and personalized disease models, a network structure formed by crosslinking of polymer biomaterials containing large amounts of water, which can provide three-dimensional structures similar to the in vivo environment. Natural, synthetic and mixed materials-based hydrogels in cell / organoid culture, in vitro tumor models, tissue engineering, immunomodulatory, and drug delivery applications.
